Proper dose spacing –Some groups want to test “human-relevant” exposure levels. On the other hand, there are recommendations for testing at doses that assure the compound is being absorbed and some toxicity will be observed. MTD is being criticized for possibly producing nonspecific effects, especially with relatively non-toxic compounds.
What is the latest thinking on proper dose spacing that is feasible and functional for regulatory assessments? When should additional dose levels (>3) be added and what dose spacing is recommended to capture toxicity AND human relevant exposures (or extrapolations).
